Pergolide is a diamine and ergoline derivative in which the beta-hydrogen at position 8 is replaced by a (methylthio)methyl group, and the hydrogen at the piperidine nitrogen (position 6) is replaced by a propyl group. It is a dopamine D2 receptor agonist with D1 and D2 agonist activity. Used as the mesylate salt in Parkinson's disease management, it was withdrawn from U.S. and Canadian markets in 2007 due to increased risk of cardiac valve dysfunction. It functions as an antiparkinson drug and dopamine agonist. It is also an organic heterotetracyclic compound, a methyl sulfide, and a conjugate base of pergolide(1+).
