Temozolomide is an imidazotetrazine with the structure of 3,4-dihydroimidazo[5,1-d][1,2,3,5]tetrazine substituted at positions 3, 4, and 8 by methyl, oxo, and carboxamide groups, respectively. It is a prodrug of MTIC, formed by spontaneous hydrolysis in the body, and functions as an oral alkylating agent. Used in the treatment of newly diagnosed malignant glioblastoma multiforme (with radiotherapy) and malignant melanoma, it serves as an antineoplastic agent, prodrug, and alkylating agent. It is also classified as an imidazotetrazine, monocarboxylic acid amide, and triazene derivative.
