Fludarabine phosphate is a purine arabinonucleoside monophosphate with 2-fluoroadenine as its nucleobase. As a prodrug, it is rapidly dephosphorylated to 2-fluoro-ara-A, which is then phosphorylated intracellularly by deoxycytidine kinase to form the active triphosphate, 2-fluoro-ara-ATP. This compound functions as a DNA chain terminator once incorporated into DNA. It is used to treat adult patients with B-cell chronic lymphocytic leukemia (CLL) who have not responded to or whose disease has progressed during treatment with at least one standard alkylating-agent-containing regimen. It acts as an antimetabolite, antineoplastic agent, immunosuppressive agent, antiviral agent, prodrug, and DNA synthesis inhibitor. It is also an organofluorine compound, a nucleoside analogue, and a purine arabinonucleoside monophosphate, and is functionally related to 2-fluoroadenine.
