Pemetrexed is an N-acylglutamic acid with the N-acyl group as 4-[2-(2-amino-4-oxo-4,7-dihydro-1H-pyrrolo[2,3-d]pyrimidin-5-yl)ethyl]benzoyl. It inhibits thymidylate synthase (EC 2.1.1.45), dihydrofolate reductase (EC 1.5.1.3), and glycinamide ribonucleotide formyltransferase (EC 2.1.2.2). It functions as an antineoplastic agent, antimetabolite, and pyrrolopyrimidine, and is a N-acyl-L-glutamic acid. It is also the conjugate acid of pemetrexed(2-).
