Momelotinib is a benzamide formed by the condensation of the carboxy group of 4-{2-[4-(morpholin-4-yl)anilino]pyrimidin-4-yl}benzoic acid with the primary amino group of aminoacetonitrile. It is an ATP-competitive inhibitor of JAK1 and JAK2 with IC50 values of 11 nM and 18 nM, respectively. It is used in the treatment of intermediate- or high-risk myelofibrosis. Momelotinib functions as an EC 2.7.10.2 inhibitor, an antineoplastic agent, an anti-anaemic agent, and an apoptosis inducer. It belongs to the classes of aminopyrimidines, morpholines, secondary and tertiary amino compounds, benzamides, and nitriles.
