Brequinar is a quinolinemonocarboxylic acid substituted with 2'-fluoro[1,1'-biphenyl]-4-yl at position 2, methyl at position 3, carboxy at position 4, and fluoro at position 6. It inhibits dihydroorotate dehydrogenase, an enzyme essential for de novo pyrimidine biosynthesis. Brequinar exhibits antineoplastic, antiviral, and immunosuppressive properties. It functions as an EC 1.3.5.2 inhibitor, an anticoronaviral agent, a pyrimidine synthesis inhibitor, and an antimetabolite. It belongs to biphenyls, monofluorobenzenes, quinolinemonocarboxylic acids, and monocarboxylic acids. It is the conjugate acid of brequinar(1-).
