Celastrol is a pentacyclic triterpenoid and monocarboxylic acid, specifically 24,25,26-trinoroleana-1(10),3,5,7-tetraen-29-oic acid with an oxo group at position 2, a hydroxy group at position 3, and methyl groups at positions 9 and 13. It functions as an antioxidant and anti-inflammatory agent, potently inhibiting mitochondrial lipid peroxidation and TNF-alpha-induced NFκB activation. It also inhibits topoisomerase II activity in vitro (IC50 = 7.41 μM) and acts as an EC 5.99.1.3 [DNA topoisomerase (ATP-hydrolysing)] inhibitor, antineoplastic agent, Hsp90 inhibitor, and metabolite.
