Cyproheptadine is formed by the oxidative coupling of position 5 of 5H-dibenzo[a,d]cycloheptene with position 4 of 1-methylpiperidine, creating a double bond between the two fragments. It is a sedating antihistamine with antimuscarinic and calcium-channel blocking properties. Used primarily as the hydrochloride sesquihydrate, it treats allergic conditions such as rhinitis, conjunctivitis, urticaria, angioedema, and pruritic skin disorders. Unlike other antihistamines, it also acts as a serotonin receptor antagonist, making it effective for vascular headaches and anorexia. It functions as a H1-receptor antagonist, serotonergic antagonist, antipruritic, anti-allergic, and gastrointestinal drug. It belongs to the piperidine class and is a tertiary amine.
