Glasdegib is a benzimidazole containing a (2R,4S)-4-{[(4-cyanophenyl)carbamoyl]amino}-1-methylpiperidin-2-yl group at position 2. It is a SMO receptor antagonist and Hedgehog signaling pathway inhibitor with an IC50 of 5 nM. Used in combination with low-dose cytarabine, it treats newly-diagnosed AML in adults aged >=75 years or with contraindications to standard chemotherapy.
